Is it possible to configure 'Self Collection' as one of the options for delivery?

Yes. You can rename and use one of the merchant configured options (like ground in Flat Shipping) by going to Preferences > Store Text > Shipping.

How do I configure if 'Self Collection' option is chosen, shipping charges are not charged ? I am using Flat Shipping option based on certain amount now.
For e.g. all orders below $50 have $10 shipping charge applied.
So if shopper selects self collection option I do not want to charge $10 even if the product sub total is less than $50

Are you using the shipping by weight table?

All ShopSite merchant configured shipping tables (i.e. Flat shipping, shipping by weight, shipping by currency) allow you to set up multiple shipping options. The default ones are Ground, 2nd Day Air, and Next Day. You can check the box to display one of the shipping options you aren't using, set it to $0 and then rename it to "Self Collection".
