Is there anyway to give a discount to preferred customers... such as club
members on a permanent bases.

Sure. With ShopSite Pro you can enable customer registration and assign registered members to a specific group. This group can also be assigned an automatic coupon (like 10% off) so that when they sign-in, the automatic coupon gets applied to their cart.

would all customers have to register or only those who wish to join the

You can make customer registration optional, as it would only have to be used by your preferred club members. If this is an exclusive club, you could even hide the sign-in link and make that a hidden URL to sign-in.

Before Customer Registration, I used the Coupon Code feature. Customers
were given their Coupon Code and placed the orders with the discount
being taken at check out. One of the benefits when I use this method is
that I can edit the Coupon to not include certain products!
