ShopSite 10 implements PayPal Payflow Pro's latest v4 programming interface (their older interfaces won't work after September 1st 2009, so this was necessary). So this may be a difference in the way their v4 interface works vs. the older v2 interface.
First, make sure you have the latest version of ShopSite 10 (I believe maintenance release r7 is currently the latest), then in Commerce Setup -> Payment -> Configure Processor make sure you have the 'Display Error Details' checkbox checked, then place a test order with a real card and don't enter your CVV2 and see what happens. If the order is rejected, you should see the detailed error message and codes returned by Payflow Pro beneath the generic error message.
